Real-time tracking of microglia in the mouse using c-fms-eGFP.
Microglia are the principal immune effector cells in the brain. They respond to insults and pathological conditions and thus contribute to tissue repair or neurodegeneration. However, the detailed kinetic and spatial relationship between microglia and neural network activity is poorly understood. We generated transgenic mice that express enhanced green fluorescent protein (eGFP) under the control of a microglia-specific promoter (c-fms). We found that c-fms-eGFP expression is developmentally regulated and limited to microglia in the brain. Furthermore, we combined the c-fms-eGFP transgenic mice with calcium imaging to track the in vivo kinetics of microglia during physiological neural activity. When activated by sensory stimulation, microglia in the somatosensory cortex increase their intracellular calcium activity within several minutes. This time course is compatible with the kinetics of sensory-evoked local field potentials as recorded electrophysiologically, suggesting that the direct observation of microglia activity may provide insight into the time course of interneuron-microglia signaling.
